Avemar (Fermented Wheat Germ Extract & Humet (Humic Acid)

# C118 Chemoprevention of in vitro cytotoxicity (MTT-test) caused by environmental mutagens (nickel, cobalt, and xenooestogens).

Aims:

The main purpose of the adjuvant application of Avemar® (a fermented wheat germ extract) and Humet® (humic acid) in the treatment of in vitro cell lines and primary cultures of granulosa and Leydig cells and MCF-7 tumor cell line was to reduce the hormonal disrupting and toxic side-effects of NiCl2, CoCl2 and Atrasin, Alaclor, Acetaclor.

We have studied the in vitro effects of Avemar® and Humet® in different experiments.

Methods:

These tests were performed on primary culture of mouse Leydig cells, human granulosa cells, HepG2 (human hepatocarcinoma),Vero (Green Monkey kidney fibroblast), and on MCF7 (human mammary adenocarcinoma ) cell line.

In vitro MTT assayIncubation with MTT was in the last 3h of cell cultivation. Determination of cytotoxicity was carried out by photometry. Determination of hormone production was made by ELISA. Experiments1) NiCl2 was used for 24h treatment on granulosa and Leydig cells and different tumor cell lines and measured the hormon production by ELISA and cytotoxicity by MTT test. 2)

These experiments were repeated in the presence of Avemar® and Humet® to prevent these toxic effects. 3) The same protocol was used for normal and transformed tumor cell lines.

Results:

Avemar® and Humet® alone had no toxic effect in our in vitro systems. The cytotoxic effect of NiCl2, CoCl2, Atrasin, Alaclor and Acetaclor has decreased at the same time by in vitro Avemar® and Humet® treatment in all cells including the MCF7 mammary tumor cell line.

Conclusion:

Our results confirmed the previous hypothesis that due to the chemoprevention by Avemar® and Humet® the toxicity and hormonal disrupting effect of different environmental pollutants were markedly reduced.

These results suggest that there is a new possibility to introduce these non-toxic chemopreventing agents in food supplements for those high risk group of people who live in polluted region.
